However, beer spoilage organisms gain resistance to iso-?ñ-acids through several hop-resistance genes. The hop-resistance gene HorA is a membrane-bound efflux pump that shows similarity to other multidrug transporters that are known for having a very wide specificity to the types of compounds that they can transport. Because breweries must go through extensive efforts sanitizing equipment to prevent spoilage, it is hypothesized that organisms expressing HorA are more resistant to industrial sanitizers than organisms that do not. I plan to clone HorA from the beer-spoilage organism Lactobacillus brevis BSO 464 into the non-hop-resistant organism Escherichia coli DH5?ñ. A minimum concentration assay will then be used to test the sensitivity of the E. coli with the cloned HorA to various sanitizing compounds including ethanol, isopropanol, a peroxide-based compound, bleach, peracetic acid, and iodine. The MIC assay will decide the appropriate concentration range to use for a dose-response assay. The colony forming units of the dose-response assay will determine the cells' percent survivorship across each compound's concentration range, which will be graphed into a Dose-Response Curve. Finally, the IC50 values (the concentration at which there is 50% survivorship) will be compared between the E. coli with and without HorA for each
