Computational docking has shown that NPC2 is able to effectively bind to steroids such as estradiol,\nsuggesting a role in endocrine disruptions. In this work, NPC2 is expressed in E. coli cell culture and isolated using differential\nsolubilization followed by affinity chromatography. Purity at each stage was analyzed via SDS-PAGE to ascertain progress in\nelimination of contaminants. Following full purification, cholesterol-binding activity was assessed using biolayer interferometry\n(BLI), alongside optimization of sterol-binding conditions through systematic variation of buffer compositions and protein\nconcentrations.
